data-widget_type=\"text-editor.default\">\n\t\t\t\t\t\t\t\t\t<p>A pandemia da covid-19 foi controlada, mas as variantes do v\u00edrus Sars-CoV-2 ainda preocupam os pesquisadores. Ainda na busca por novos imunizantes, um grupo de pesquisadores do Instituto Ci\u00eancias Biol\u00f3gicas (ICB) da UFMG desenvolveu uma plataforma de nanopart\u00edculas lip\u00eddicas para identificar imunizantes eficazes contra variantes da infec\u00e7\u00e3o.\u00a0<\/p><p>Os passos do estudo que levou ao desenvolvimento dessa plataforma est\u00e3o detalhados no artigo\u00a0<em>Nanoparticle-based DNA vaccine protects against Sars-CoV-2 variants in female preclinical models<\/em>, publicado no m\u00eas passado na revista Nature Communications. A tecnologia empregada pelo grupo \u00e9 diferente da utilizada nas vacinas de RNA. \u201cApesar da semelhan\u00e7a com as vacinas de RNA j\u00e1 aprovadas, nossa abordagem \u00e9 diferente porque utiliza DNA encapsulado em nanopart\u00edculas lip\u00eddicas&#8221;, explica o professor Pedro Pires Goulart Guimar\u00e3es, do Departamento de Fisiologia e Biof\u00edsica do ICB e coordenador do estudo.<\/p><p>O grupo de pesquisa j\u00e1 concluiu os testes pr\u00e9-cl\u00ednicos da vacina, incluindo a avalia\u00e7\u00e3o de\u00a0sua efic\u00e1cia e imunogenicidade em camundongos e hamsters. Lays Cordeiro Guimar\u00e3es, doutoranda respons\u00e1vel pela pesquisa, explica o processo de desenvolvimento da plataforma, comparando-a a uma biblioteca de nanopart\u00edculas, em que\u00a0cada nanoformula\u00e7\u00e3o carrega um c\u00f3digo de barras de DNA distinto.\u00a0<\/p><p><strong>Mistura de nanoformula\u00e7\u00f5es<br \/><\/strong>\u201cPreparamos uma mistura de todas as nanoformula\u00e7\u00f5es da biblioteca e injetamos em um mesmo animal e, por meio de equipamentos e t\u00e9cnicas, conseguimos identificar qual a melhor formula\u00e7\u00e3o para entrega de DNA em cada parte do organismo\u201d, explica. Ap\u00f3s sele\u00e7\u00e3o criteriosa, a melhor formula\u00e7\u00e3o para entrega de DNA em \u00f3rg\u00e3os linfoides foi identificada e utilizada para encapsular o DNA que codifica a prote\u00edna spike do v\u00edrus da covid-19.<\/p><p>Os resultados dos testes demonstraram resposta imunol\u00f3gica eficaz nos animais que receberam a vacina e s\u00e3o\u00a0compar\u00e1veis aos da vacina da Pfizer\/BioNTech, que utiliza a tecnologia de RNA. \u201cOs resultados foram semelhantes nos grupos que usaram a nossa vacina e naqueles imunizados com a vacina de RNA da Pfizer\/BioNTech. As\u00a0redu\u00e7\u00f5es\u00a0de les\u00e3o pulmonar, carga viral e letalidade foram semelhantes em ambos os grupos&#8221;, explica Lays. Os resultados observados nos grupos imunizados com vacina de DNA s\u00e3o promissores, especialmente considerando os desafios apresentados pelas variantes do v\u00edrus Sars-CoV-2.<\/p><p>O trabalho reuniu os\u00a0pesquisadores Frederic Fr\u00e9zard, Mauro Martins Teixeira, Vivian Vasconcelos Costa e Renato Santana de Aguiar, todos\u00a0do ICB. Para a pesquisadora, tal plataforma tecnol\u00f3gica pode ser adaptada para pesquisas de imunizantes contra outras doen\u00e7as. \u201cA chikungunya, por exemplo,\u00a0pode deixar sequelas graves, e ainda\u00a0n\u00e3o existe vacina contra essa doen\u00e7a no Brasil. Uma parte do nosso\u00a0grupo est\u00e1 fazendo os testes para a chikungunya, e \u00e9 poss\u00edvel que essa tecnologia seja testada para muitas outras doen\u00e7as.\u201d<\/p><p>O trabalho que culminou no artigo publicado na Nature Communications foi desenvolvido no\u00a0<a href=\"https:\/\/ufmg.br\/comunicacao\/noticias\/laboratorio-de-biosseguranca-nivel-3-do-icb-comeca-a-funcionar-em-fevereiro\">Laborat\u00f3rio de Biosseguran\u00e7a N\u00edvel 3 (NB3) do ICB<\/a>. A instala\u00e7\u00e3o, que se destina a pesquisas com v\u00edrus, bact\u00e9rias, fungos e outros microrganismos causadores de doen\u00e7as transmitidas pelo ar,\u00a0entrou em opera\u00e7\u00e3o em janeiro de 2022.
